What is DSIP? A sleep-associated nonapeptide

DSIP is the nine-amino-acid peptide Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, originally identified in cerebral venous blood collected from rabbits during electrical stimulation of the thalamus that induced sleep. The peptide's specific mechanism of action remains an open research question — despite decades of investigation, no dedicated 'DSIP receptor' has been definitively characterised — which has made it a persistent object of study in the neuropeptide literature.

DSIP mechanism and research pathways

Published research has documented DSIP's association with slow-wave (delta) sleep architecture in EEG studies, modulation of the hypothalamic-pituitary-adrenal (HPA) axis, protective effects against oxidative stress in neuronal cell models, modulation of somatostatin and beta-endorphin release, and anticonvulsant activity in seizure models. The absence of an identified receptor suggests DSIP may act through multiple upstream neuroendocrine pathways rather than a single defined signalling target.

Reconstitution and stability

Store the lyophilised vial at -20°C, sealed and protected from light. Reconstitute with bacteriostatic water (0.9% benzyl alcohol) — typically 1ml per 5mg vial. Add solvent slowly down the side of the vial; do not shake. Refrigerate the reconstituted solution at 2–8°C and use within 28 days. Do not refreeze.
